Only the pluripotent cells of the E3.5 mouse blastocyst express the Oct-4 transcription factor (green); the trophectoderm lacks expression of this lineage marker (red, DNA staining by propidium iodide). At 4 days in vitro, mouse blastocysts form characteristic outgrowths in which pluripotent cells continue to express Oct-4 (green); other cell lines e.g. the primitive endoderm or trophectoderm lack this factor (DNA red).
